Abstract
The accumulation of protein aggregates is the pathological hallmark of many devastating proteopathic neurodegenerative diseases (PNDs), including Alzheimer´s and Parkinson´s diseases (AD and PD). Despite a common origin in the accumulation of specific proteins, the clinical and pathologic phenotype of these diseases exhibit conspicuous variability within and among patients. These heterogeneous phenotypes have been anticipated to occur due to the existence of distinct protein aggregate morphotypes.
For over a decade, we have developed multimodal thiophene-based ligands that can be used for optical assignment of distinct protein aggregates and in this talk, I will show how these ligands have evolved to become an outstanding method for assessing protein aggregate polymorphism in PNDs. Overall, the chemical design of novel ligands for ex vivo and in vivo detection of protein aggregates by a variety of fluorescent modes, such as hyperspectral- and fluorescence life-time imaging, will be discussed and exemplified by multidisciplinary studies that have generated novel molecular insights regarding the underlying pathogenesis of PNDs. In addition, the chemical evolution of these ligands towards clinical diagnostic and therapeutic use will be presented. Finally, the concept of applying multimodal thiophene-based ligands within other research areas, such as cancer stem cells and bacterial infection will be demonstrated
