Abstract
Sleep apnea (SA) is a highly prevalent respiratory disorder affecting several million adult men and women.
SA is characterized by periodic cessation of breathing during sleep, either due to obstruction of the upper airway (obstructive sleep apnea, OSA) or disruption of the respiratory rhythm generation by the central nervous system (central sleep apnea, CSA). Current treatment strategies include Continuous positive airway pressure (CPAP) for OSA and Adaptive servo-ventilation for CSA. However, Many OSA patients do not respond to CPAP or find CPAP difficult to tolerate. Adaptive ventilation in heart failure patients was associated with 30% mortality with no demonstrable benefits for CSA. Hence, there is an unmet clinical need to develop alternative therapies for treating SA.
Here, Dr. Prabhakar will present: a) pre-clinical murine model exhibiting OSA and CSA and b) rational drug design targeting chemoreceptor loop gain and hypoglossal tone to treat SA.
