Even seemingly identical cells show great non-genetic transcriptome variation. These subtle differences originate from small fluctuations in transcriptional and post-transcriptional regulators as well as chromatin structure. Excitingly, even complex cell features such as cell lineage relationships and micro-environments leave discernable footprints on the transcriptome. These signatures therefore allow to infer proxies for these features, adding additional information to single-cell RNA-sequencing and other single-cell gene expression datasets, enabling exciting applications especially in the context of intra-tumor heterogeneity.
